DUSP6在两种不同多能干细胞中的调控作用研究

摘要 目的探讨DUSP6的同一个调控区域在2种不同多能干细胞中的调控作用。方法采用亚克隆、实时荧光定量聚合酶链反应(PCR)、双荧光素酶活性检测系统及神经干细胞诱导等方法来确定DUSP6在胚胎干细胞未分化状态中起调控作用的区域,在神经干细胞中同样有活性表达。结果 DUSP6同一个调控区域在胚胎干细胞和神经干细胞未分化状态中具有活性表达,而其活性在分化状态下减弱或消失。结论 DUSP6同一个调控区域(82bp)在胚胎干细胞和神经干细胞的多能性阶段具有活性表达。 Objective To study DUSP6 expression in undifferentiated state of embryonic stem (ES) cells and neural stem (NS) ceils. Methods Using subclone, real-time PCR, dual-luciferase reporter assay and neuro- genesis, we demonstrate that regulatory region of DUSP6 shows its activity in multi-potent ES cells and neural stem cells. Results Overexpression of pluripotent state-specific of DUSP6 in undifferentiated state of ES cells and NS cells, thus, shows weak activity in two distinct differentiated stem ceils. Conclusion The identified regulatory element (82bp) of DUSP6 is specifically expressed in two distinct pluripotent stem ceils.
