清肠健脾愈疡汤对湿热型溃疡性结肠炎大鼠MIF、IL-6的影响

Experimental Study of Decoction of Qing-chang Jian-pi Yu-yang on Ulcerative Colitis Rat with Type of Humid Heat

目的:研究清肠健脾愈疡汤对湿热型溃疡性结肠炎(UC)大鼠MIF、IL-6的影响。方法:将雄性SD大鼠60只,随机分为6组。采用高脂饮食、白酒加湿热环境干预结合2,4,6-三硝基苯磺酸诱导法复制湿热型UC模型,模型复制后给药2周,检测大鼠结肠黏膜MIF表达含量及血清IL-6含量。结果:与模型组比较,柳氮磺胺吡啶组、清肠健脾愈疡汤灌肠组、灌胃组以及灌肠加灌胃组大鼠MIF表达及IL-6的参数显著降低(P<0.01);与SASP组比较,中药灌肠组(P<0.05)、灌肠加灌胃组(P<0.01)大鼠MIF表达参数含量显著降低,中药灌胃组(P<0.05)、清肠健脾愈疡汤灌肠组(P<0.01)、灌肠加灌胃组(P<0.01)大鼠IL-6的参数显著降低;清肠健脾愈疡汤3组中,灌肠加灌胃组大鼠MIF表达及IL-6的参数显著降低(P<0.01)。结论:清肠健脾愈疡汤治疗湿热型UC的作用机制之一是抑制MIF的表达,进而下调促炎细胞因子IL-6,减轻炎症反应,促进溃疡愈合。且不同的给药方法治疗效果也有差异。

Objective ： To observe the effect of Decoction of Qing-chang Jian-pi Yu-yang on MIF and IL-6 in rat with ulcerative colitis （UC）. Methods：60 male SD rats were divided into six groups randomly. The models of UC with type of humid heat were made with high fat diet and distillate spirit combined with 2,4,6-TNBS （trinitro-ben- zene-sulfonic acid） in wet heat surroundings. After 2 weeks MIF and IL-6 were detected. Results ： Compered with in model control group, MIF and IL-6 were decreased markedly in SASP group, treatment group 1 （with coloclysis）, treatment group 2 （with intragastric administration） and treatment group 3 （with coloclysis and intragastrie admin- istration） （ P 〈 0. 01 ）. Compered with in SASP group, MIF was decreased markedly in treatment group 1 （ with coloclysis） （ P 〈 0. 05 ） and treatment group 3 （ with coloclysis and intragastric administration） （ P 〈 0. 01 ）, and IL-6 was also decreased in treatment group 2 （ with intragastric administration） （ P 〈 0. 05 ）, treatment group 1 （ with coloclysis） （P 〈 0.01 ） and treatment group 3 （with coloclysis and intragastric administration） （P 〈 0. 01 ）. Among the three treatment group MIF and IL-6 were decreased the most in treatment group 3 （with coloclysis and in- tragastric administration） （P 〈 0. 01 ）. Conclusion：One of the mechanisms of action of Decoction of Qing-chang Jian- pi Yu-yang with type of humid heat is suppressing the expression of MIF, and then regulating IL-6, so as to mitigate inflammatory reaction and promote ulcer recovery. And different medication has different curative effect.
