摘要 目的探讨TRAIL基因第五外显子3'-UTR区1525G/A、1595C/T与特发性血小板减少性紫癜的相关性。方法采用病例对照的方法,选择ITP患者132例,正常对照者130例,提取外周血基因组DNA,采用PCR-RFLP法分析TRAIL基因1525G/A、1595C/T位点的基因型,计算基因型和等位基因频率,进行χ2检验。结果TRAIL基因第五外显子3'-UTR区1525G、1595C等位基因频率ITP组高于正常对照组,但是两组比较1525G/A、1595C/T位点基因型分布无统计学差异(P=0.367),两组间等位基因频率比较无统计学差异(P=0.262)。结论TRAIL基因第五外显子3'-UTR区1525G/A、1595C/T位点基因多态与ITP的遗传易感性没有相关性。 Objective To explore the association of TRAIL polymorphism with idiopathic thrombocytopenic purpura(ITP) in Chinese Han nationality from Shandong province.Methods This study followed a case-control design,132 patients with ITP and 130 healthy controls were included.Genomic DNA were extracted from peripheral blood,and the polymorphism of TRAIL gene at 1525G/A and 1595C/T were detected by polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP),while genotype and allele were analyzed with Chi-square test.Results Frequencies of 1525G/1595C allele in ITP group were higher than those in healthy controls,but no significant difference in distribution of genotype was shown between ITP group and healthy controls(P=0.367).No significant difference in distribution of allele was shown between ITP group and healthy controls(P=0.262).Conclusion Polymorphisms of TRAIL gene 1525G/A,1595C/T positions in 3′-UTR of exon 5 weren′t associated with susceptibility of ITP.
