产碱性木聚糖酶菌株的选育及其酶解产物鉴定 被引量:2

Mutation breeding of alkaline xylanase-producing strains and identification of enzymic hydrolysates

摘要 以实验室保藏的1株产碱性木聚糖酶的坎皮纳斯类芽孢杆菌(P.campinasensis)xy-7为出发菌株,经紫外线、硫酸二乙酯单一及复合诱变处理,选育获得1株遗传性能稳定的高产突变株DU-36,酶活达到322IU/mL,比出发菌株提高了1倍。采用纸层析法分析,结果表明不同反应时间下酶解产物均为木二糖,推断该木聚糖酶为外切型木聚糖酶。 A mutant DU-36 with high alkaline xylanase activity was obtained after the strain P.campinasensis xy-7 was treated by UV-ray and diethyl sulfate. The enzyme activity reached 322 IU/mL, which increased by 100%. The result by paper chromatography indicated that the xylanase hydrolysate was always xylobiose under different hydrolysis times. From this reason, the alkaline xylanase produced by DU-36 belonged to excision enzyme.
