摘要
以琼脂中的冷溶性琼脂多糖为研究对象,探讨其体内抗肿瘤作用及机理。10~15℃下水浸提琼脂中的多糖,并对其进行了组分分析。小鼠腋下接种H22肝癌细胞建立动物模型,以1000mg/kg·d灌胃给药。通过瘤重、抑瘤率、脏器指数、免疫器官切片HE染色、脾细胞增殖能力、NK细胞杀伤活性测定、吞噬细胞吞噬能力测定等研究其抗肿瘤作用和机理。测得其总糖含量为76.74%,抑瘤率为77.81%,小鼠免疫系统得到保护,增强了抗肿瘤作用。冷溶性琼脂多糖有显著的抗肿瘤作用,在保护免疫系统,提高免疫力方面有很好的作用。
To study the antitumor activity in vivo of low temperature dissolved agar polysaccharide. Low temperature dissolved agar polysaccharide was extracted with water from agar at 10-15 %. The isolate was analyzed that it contained 76.74% polysaccharide. H22 tumor bearing mice were established and treated by gavage with 1000 mg/kg of the polysaccharide. The tumor weight, tumor inhibition rate, thymus index, HE staining of thymus, lymphocyte proliferation, kill rate of NK cells, detection of phagocytic were measured to study antitumor function in vivo. The low temperature dissolved agar polysaccharide had definite antitumor effect and the tumor inhibition rate is 77.81%. It could protect the immune system and promote the immune function of tumor bearing mice.
