慢性非细菌性前列腺炎大鼠勃起功能的观察 被引量:3

The erectile function of rats with chronic nonbacterial prostatitis

摘要 目的观察慢性非细菌性前列腺炎(CNP)大鼠的勃起功能。方法取4个月龄雄性SD大鼠10只,在无菌条件下取出前列腺组织,制作前列腺组织蛋白提纯液(20g/L)。另取2个月龄SD大鼠50只,随机分为CNP组和对照组,各25只。CNP组于第0、30天皮内多点注射前列腺组织蛋白提纯液与弗氏完全佐剂的等体积混悬液1.0ml,同时腹腔注射百白破疫苗0.5ml;对照组同法注射等量生理盐水。第45天通过电刺激法来测定两组大鼠的勃起功能,以电刺激阴茎海绵体神经时海绵体内压力的最大值与对应平均动脉压的比值(记为bR)表示大鼠的勃起功能。每只大鼠重复测定3次。勃起功能测定结束后,留取血清,顺磁性微粒化学发光免疫法检测两组大鼠的血清睾酮浓度;留取部分前列腺组织,苏木素.伊红(HE)染色,光镜下观察前列腺腺体和间质的病理变化。结果CNP组大鼠大体形态可见前列腺组织严重充血水肿,与周围组织粘连,结构不完整。光镜下见前列腺组织结构遭到不同程度破坏,问质和腺体内有弥漫的淋巴样组织增生和慢性炎细胞浸润。对照组大鼠无上述炎症表现。CNP组和对照组勃起功能指标bR总的平均值分别为(68.07±10.54)%和(70.44±11.18)%,差异无统计学意义(P〉0.05)。两组的血清睾酮浓度分别为(3.84±1.50)、(4.75±1.75)ug/L,差异亦无统计学意义(P〉0.05)。结论CNP大鼠的勃起功能与正常大鼠比较无明显降低,推测临床上慢性前列腺炎患者可能并不存在器质性勃起功能障碍。 Objective To study the erectile function of rats with chronic nonbacterial prostatitis (CNP). Methods The prostate tissues of 10 male SD rats (4 months old) were taken out under aseptic condition, and the prostate tissue homogenate supernate (20 g/L) was made. Another 50 male SD rats (2 months old) were randomly divided into CNP group and control group, with 25 rats each group. Each rat of CNP group was immunized with 1.0 mL prostate tissue homogenate supernate emulsified by isopyknic Freund' s complete adjuvant intradermally in the multiple points, and simultaneously immunized with 0. 5 mL pertussis diphtheria tetanus vaccine intraperitoneally on the 0 and 30th day. Each rat in control group was injected with equivalent normal saline in the same way. On the 45th day, the erectile function of each rat in two groups was evaluated by the ratio of maximum intracavernous pressure/mean arterial pressure (bR) during electrostimulation of the cavernous nerve. Each rat was measured for three times. After the measurement of erectile function, the blood samples were obtained by cardiac puncture and the levels of serum testosteroncs were determined by paramagnetism chemiluminescence immunoassay. Besides, the prostatic tissues were harvested and examined for the histopathological characteristics. Results In CNP group, prostate tissues showed serious congestion and edema, adhesion with surrounding tissues, and less integrity of structure, macropathologically. Histologically, the prostatic tissues were characterized by lymphoid tissue proliferation and chronic inflammatory cell infiltration in the stromal connective tissue around the acini or ducts. In control group, no inflammatory infiltration as above was seen. The mean bR in CNP and control groups was (68.07 -+ 10. 54) % and (70. 44 -+ 11.18 ) %, respectively, with no significant difference ( P 〉 0. 05). The average serum levels of testosterones in CNP and control groups were (3. 84-+ 1.50) ug/L and (4. 75 -+ 1.75 ) ug/L, respectively, with no significant difference ( P 〉 0. 05 ). Conclusion Compared to the normal rats, the erectile function in CNP rats was not significantly decreased, suggesting that there seems to be no organic erectile dysfunction in the patients with chronic prostatitis.
