摘要
目的观察间隙连接蛋白40(Cx40)在培养新生大鼠心房肌细胞中的表达变化。方法对6个实验组36个培养皿,在培养第2、7、14、21、28、35天,用免疫细胞化学方法进行Cx40的测定。结果培养第2天心房肌细胞开始表达Cx40,表达部位主要在细胞膜及细胞质中,呈点状分布,细胞间阳性表达率为(6.2±0.4)%;第7天细胞膜和细胞质中点状分布减少,表达主要集中在细胞膜连接处,细胞间阳性表达率(82.6±7.2)%;第14天侧-侧连接处、端-端连接处均有表达,细胞膜未见表达;第21天闰盘处表达明显,并呈条状分布,阳性表达率(90.1±8.9)%;第28天时Cx40颗粒出现紊乱分布,阳性表达率为(79.8±3.5)%;第35天侧闰盘处、端闰盘处及侧一端闰盘处阳性表达率为(49.4±5.7)%。结论Cx40在培养的心房肌细胞中的表达随培养时问变化,与心房肌细胞功能相关。
Objective To investigate the expression profiles of eonnexin40 (Cx40) in cultured atrial muscles of newborn rats. Methods These cells were divided randomly into 36 culture dishes and 6 experiment groups. The expression of Cx40 was detected by using immunocytoehemistry on the 2nd, 7th, 14th, 21st, 28th, and 35th day respectively. Results On the 2nd day after culture, Cx40 was expressed in the cardiac myocytes, mainly localized on the cell membrane and in the cytoplasma, and the positive expression rate in cell-cell was (6. 2 ±0. 4)%. On the 7th day, the positive expression rate was (82. 6 ± 7. 2)% , the expression of Cx40 on the cell membrane and in the cytoplasma was decreased and mainly localized between the junction of cell membrane. On the 14th day, at the junction of both side-side and endend, Cx40 were expressed, but no expression on the cell membrane was detected. On the 21st day, the positive expression rate was (90. 1 ± 8. 9 )% , and Cx40 was expressed at intercalary disc and distributed like strip. On the 28th day, the positive expression rate was (79.8 ±3.5)% , Cx40 was expressed disorderly. On the 35th day, the positive expression rate was (49. 4 ± 5.7 ) %, and Cx40 was expressed in side-intercalary disc (ID), end-ID and side-end ID. Conclusion The expression of Cx40 in the cultured atrial muscle ceils of neonatal rats was concerned with the atrial muscle cellular function and changed with the cultural time.
