摘要 对一株裂褶菌(Schizorhyllum commune ATCC38548)进行液态发酵生产纤维素酶。通过单因素试验考察发酵时间、碳源、碳源质量浓度和接种量等因素对菌株产酶的影响,用二硝基水杨酸法(DNS法)对羧甲基纤维素酶(CMC)、滤纸酶(FPA)和微晶纤维素酶(AVI)进行酶活测定。结果表明:3种酶的分泌高峰不一致,但在第6天时纤维素酶总酶活达到最高。滤纸是诱导裂褶菌产纤维素酶的良好碳源,且质量浓度为20g/L时产酶效果最佳;接种量为5mL时,总酶活相对最高。不同的培养条件对裂褶菌产CMC、FPA和AVI的影响各异。 A strain called Schizorhyllum commune ATCC38548 was cultivated in submerged state to produce cellulases.The effects of different factors,such as fermentation time,carbon sources,mass concentration of carbon sources and inoculation volumes,were investigated by single-factor experiments.Initrosalicylic acid colorimetric method(DNS) was used to measure the activities of carboxymethyl cellulase(CMC),filter paperlyase(FPA) and crystallite cellulase(AVI).The results showed that production summits of 3 enzymes was inconsistent,but the total activities of cellulase had a maximum value after 6 days fermentation.Filter paper was a good carbon source due to its inductivity for cellulase,and 20g/L of mass concentration and 5mL of inoculation volume brought the best output of cellulase.All these 3 enzymes had different activities in different culture conditions.
