钙调神经磷酸酶催化结构域的活性和性质 被引量:2

摘要 钙调神经磷酸酶(Calcineurin,CN)是唯一的Ca2+/钙调素(Camodulin,CaM)依赖的蛋白磷酸酶,由催化亚基A(CNA, 61 ku)和调节亚基B(CNB, 19 ku) 1:1组成.用 PCR方法构建了仅含CNA催化结构域的编码区cDNA序列.经在E.coli中诱导表达,纯化,制备了CNA催化结构域多肽.以PNPP为底物,研究了其磷酸酶活力.结果表明,在有CaM和CNB时,其活性为CNA活性的20倍.CNA催化结构域多肽的最适反应温度为40℃,其最适反应pH值为8.0.不同离子对其活性的影响为: Mn2+仍为其强激活剂,但 CNA催化结构域多肽不再受 Ca2+的调节.在 6 mmol/LMg2+存在下,加Ca2+或EGTA,催化结构域多肽的活性相同.
