摘要
以玛丽安万年青( Dieffenbachia Tropic Marianne) 的顶芽或侧芽茎尖作外植体,通过二次表面灭菌,得到茎尖无菌培养物.根据茎尖愈伤组织诱导率和生长速度2 项指标,筛选出玛丽安万年青茎尖离体培养的培养基.MS+ BA4 .Omg/L 的固体培养基适宜于诱导茎尖形成愈伤组织和愈伤组织生长.愈伤组织转入MS+ BA2-Omg/L 的固体培养基上分化形成丛生芽.从愈伤组织上切取1cm 以上的芽转入MS+ NAA1-Omg/L 的固体培养基上生根壮苗.试管苗壮苗培养50d 左右移栽,成活率95 % .
The explant originated from shoot apex of terminal bud or lateral bud of Dieffenbachia Tropic Marianne was throughly surface sterilized by double sterilizing.According to the frequency of induction and growth speed of callus,which were used in this experiments,the optinum medium for cultured shoot apex in vitro was MS mediums with BA 4.0 mg/L.The callus differentiated into plenty of buds on an agar-solidified MS mediums containing BA 2.0mg/L.Shoots cut from callus were rooted in an agar-solidified MS mediums containing NAA 1.0mg/L.Plantlets were transplanted after rooting culture for 50 days.The survival rate of transplanted plantlet was 95%.
