摘要
用显微操作技术将Ca2 + 特异荧光染料Calcium greenTM 1注射入GV期卵母细胞的细胞质内 ,在激光扫描共聚焦显微镜下观察生发泡破裂前Ca2 + 的时间和空间分布的变化 ,利用Lasersharp软件进行分析 ,发现一种有规律的Ca2 + 波动 ,其波动周期约为 90s .卵母细胞内Ca2 + 的分布并不均匀 ,核膜周围的Ca2 + 浓度最高 ,细胞质中次之 ,而生发泡内部Ca2 +
The fluroscence dye Calcium green TM 1 was injected into the oocytes to indicate the free Ca 2+ and a laser scanning confocal microscope was used to determine the change and the distribution of Ca 2+ before GVBD .It is found that there were many calcium oscillations after releasing from follicle.The oscillation frequency is about 90 s and continue for 2~3 h. The distribution of Ca 2+ is different between the plasma and nucleus, the Ca 2+ concentration is highest around the nuclear membrane,it is lower in plasma and is lowest inside the nucleus.
