溶血磷脂酸对小胶质细胞炎性分泌的影响 被引量:1

The effect of LPA on inflammatory secretion of microglia

摘要 目的观察溶血磷脂酸(LPA)对小胶质细胞活化及其炎性因子分泌的影响。方法小胶质细胞系BV2细胞复苏后传代培养,分为对照组和LPA组,在30 min、1、3和6 h收取上层培养基,用ELISA法测定各组培养基中IL-1β和TNF-α的浓度。结果 LPA干预后BV2细胞分泌的IL-1β和TNF-α在1 h开始明显增加,IL-1β的分泌在3 h达到最高峰而TNF-α的分泌在6 h内持续升高(P<0.01)。结论 LPA可显著促进BV2细胞IL-1β和TNF-α的分泌,在3 h时促进作用最为显著。LPA可诱导BV2细胞活化及炎性因子分泌增加,提示LPA在小胶质细胞活化及炎性因子分泌中发挥了重要作用。 Objective To observe the effect of LPA(lysophosphatidic acid) on microglia activation and inflammatory secretion.Methods Microglia cell line BV2 were randomly divided into two groups: control group and LPA group.At different time points(30 min、1 h、3 h、6 h),medium were harvested for further use.ELISA was used to detect the concentration of IL-1β and TNF-α in the medium.Results Compared with the control group,the concentration of IL-1β and TNF-α in LPA group were began significantly increased at 1 hpoints.The concentration of IL-1β reached peak lever at 3 h points and the concentration of TNF-α continue to rise in 6 h.(P<0.01).Conclusions LPA can promote microglia activation and inflammatory secretion in vitro.Our results suggested that LPA might play an important role in microglia activation and inflammatory secretion.
