摘要
目的:建立胃空肠Roux-en-Y胃旁路(RYGB)术GK大鼠实验动物模型,并初步探讨该旁路术对GK大鼠体重及血糖的影响。方法:将30只12周龄的雄性GK大鼠随机分为手术组(O组)10只、假手术饮食控制组(C组)及假手术空白对照组(B组)各10只。每日称量大鼠体重、进食量及每周测量空腹血糖,术后第1、2个月行口服糖耐量试验。结果:O组死亡2只,B组死亡1只;O组在术后2周内体重下降幅度最大,且体重上升明显慢于其他两组;O组术后空腹血糖明显低于术前及B组和C组(P<0.01);O组在术后第1、2个月糖耐量曲线下面积较其他两组缩小范围最大,差异有统计学意义(P<0.01)。结论:通过GK大鼠建立RYGB动物模型是可行的,RYGB术可有效控制GK大鼠体重和降低血糖。
Objective: To establish the experimental animal model of gastric bypasssurgery (RYGB) through the GK-rat, and to study the effect of gastric bypass surgery on weight reduction and reduce blood glucose. Methods: Male rats (30 GK-rat) were randomized into three groups:gastric bypass surgery was performed in 10 male GK-rats as the study group (0 group), sham operation was performed in 10 GK male rats which diet control after operation as the study control group (C group~ and laparotomy was performed in 10 GK male rats which were given normal diet control after operation as the blank control group (B group). Body weight and food intake were recorded before surgery and every day after surgery, FPG were recorded before surgery and every week after surgery. OGTT were recorded before surgery and first and second month after surgery. Results: The number of death was 2 in the study group, the survival rate is 80%, and the maximum weight loss was happened in the second week after surgery, Thereafter, weight rebounded slower than group B and C. The FPG under the level of before surgery, as well as group B and C obviously. Compared to that of the two groups, postoperative RYGB group showed a significant decrease. Conclusion: It is feasible to established Roux-en-Y gastric bypass surgery animal model through the GK rat. RYGB can control weight and blood glucose of GK-rst effectively.
