不同图像分析软件定量计算肌纤维类型数量及面积 被引量:3

Quantification and Size of Muscle Fibers based on different Image Analysis Software

摘要 目的:比较不同图像处理软件在定量计算不同类型肌纤维数量和面积时的优缺点,找到精确定量的简便方法。方法:采用尾悬吊模型造成大鼠肌肉去负荷萎缩后,应用ATP酶异染法对腓肠肌纤维冰冻切片进行染色分型。Image J和Image-Pro plus软件进行不同类型肌纤维的计数;Image-Pro plus和Photoshop计算不同类型肌纤维的面积百分比。结果:Image J可以精确的对不同类型肌纤维进行计数,而Image-Pro plus往往会多计数一些杂点而导致计数不准确;在对Ⅰ型肌纤维面积进行计算时,Image-Pro plus和Photoshop的结果无显著性差异;而计算Ⅱa和Ⅱb型的面积时,两种软件有显著性差异(P<0.05)。结论:使用Image J软件可以方便、准确进行不同类型肌纤维的计数;染色效果好,肌纤维类型区分明显的片子可用Image-Pro Plus快速进行面积统计;区分度不太理想的片子可以用Photoshop进行精确选择后计算面积。 Objective: Compare the advantages and disadvantages of different software in quantitatively count different fiber types,so as to find convenient and accurate method to count different fiber types.Method: Metachromatic dye-ATPase staining was used to distinguish fiber types of the gastrocnemius.The software of Image J and Image-Pro Plus was used to count the numbers of different fiber types respectively.The software Image-Pro Plus and Photoshop was used to calculate the areas of different fiber types respectively.Result: Image J can precisely count the numbers of different fiber types,while Image-Pro Plus will additionally count some noise points so that the results are not correct.There are no significant differences when calculate the areas of type Ⅰ,but significant differences(p0.05) when calculate type Ⅱa and Ⅱb,analysis with Image-Pro Plus and Photoshop.Conclusion: Image J can conveniently and accurately count the numbers of different fiber types.For good quality pictures,Image-Pro Plus can fast select and calculate the areas;but for the bad ones(especially difficult to distinguish type Ⅱa and Ⅱb),Photoshop can precisely select and calculate.
