体外培养与扩增对脐血CD34^+细胞及HLA抗原表达的影响

Effect of Cultivation and Expansion in Vitro on CD34^+ Cells and HLA Antigen Expression of Umbilical Cord Blood

采用免疫组织化学方法．观察脐血细胞体外培养与扩增前后ＣＤ３４~＋细胞的比例以及ＨＬＡ抗原的表达。结果表明：脐血ＣＤ３４~＋细胞的百分比以及ＨＬＡ抗原的表达于体外培养前后无显著性变化（Ｐ＞０．０５），而在细胞因子的联合作用下，经一定时间的扩增后，ＣＤ３４~＋细胞百分比下降（Ｐ＜０．０１），ＨＬＡ抗原的表达有一定程度的提高。提示：体外扩增对脐血ＣＤ３４~＋细胞的比例以及ＨＬＡ抗原的表达有一定的影响，而未加细胞因子的体外直接培养则不产生明显影响。

The expression of HLA antigen of umbilical cord blood (UCB) and the variations of rate of CD34^+ cells before and after cultivation in vitro and expansion were observed by using immunohistochemical mothed (APAAP). The results showed that the HLA antigen expression of UCB and the rate of CD34^+ cells had no change before and after culture in vitro (P>0. 05). But the HLA antigen expression of UCB was increased and the rate of CD34^+ cells was deceased (P<0. 01 ), after expanded by adding cytokines and HLA antigen expression was elevated to some extent. It was suggested that the expansion in vitro has an influnce on biology of UCB, but cultivation alone has no influnce on it.
