重型肝炎患者红细胞补体Ⅰ型受体基因多态性与血清可溶性补体Ⅰ型受体含量的变化及意义研究 被引量:1

The study of the change and significance of CR1 gene polymorphism and serum soluble CR1 of erythrocyte in patients with severe hepatitis

摘要 目的研究重型肝炎患者红细胞补体Ⅰ型受体(CR1)密度基因多态性的分布及血清可溶性补体Ⅰ型受体(sCR1)含量的变化。方法选择2011年3~8月在解放军第302医院住院治疗的34例重型乙型肝炎患者为研究对象,选择50例慢性肝炎患者作为对照组。采用PCR和HindⅢ酶切技术检测红细胞CR1密度基因多态性,采用ELISA定量试剂盒检测血清sCR1含量。结果重型肝炎患者红细胞CR1基因HH型的患者显著高于慢性肝炎组,二者比较差异高度显著(P<0.01);重型肝炎患者血清sCR1含量显著高于慢性肝炎(P<0.001),并与PTA变化显著负相关(r=-0.81)。结论重型肝炎患者的红细胞CR1基因型以HH为主,与慢性肝炎患者的分布存在质的差别,其发生机制可能与个体过敏体质关系密切,血清sCR1含量变化与重型肝炎患者肝脏功能损伤程度密切相关。 Objective Study on distribution of density-dependent gene polymorphism of erythrocyte complement receptor type 1(CR1)and concentrations of serum soluble complement receptor type I(sCR1) in patients with severe hepatitis.Methods 34 cases of severe hepatitis patients with hepatitis B in March-August 2011 in the PLA's 302 hospital for treatment as the object of study,50 patients with chronic hepatitis as a control group.PCR and restriction fragment length polymorphism(RFLP)technology for detecting red cell CR1 gene polymorphism and the quantitative ELISA Kit for detection of serum sCR1 concentrations had been used in this study.Results The gene of CR1 with HH in patients with severe hepatitis patients was significantly higher than the group of chronic hepatitis(P0.01).The concentrations of sCR1 in serum in patients with severe hepatitis we significantly higher than that in patients with chronic hepatitis B(P0.01),and significant negative correlation with PTA changes(r=-0.81).Conclusions The distribution of density-dependent gene polymorphism in red blood cell in patients with severe hepatitis mainly is HH type.There is archery target difference with chronic hepatitis patient's distribution of CR1 gene.The mechanism of severe hepatitis may be occurred with hypersensitivity.There is a close relationship between the sCR1 constitution and liver damage.
