青蒿琥酯对肿瘤细胞损伤修复基因表达的影响

Effects of artesunate on expression of damage repair gene in tumor cells

摘要 目的运用逆转录聚合酶链反应(RT-PCR)技术研究青蒿琥酯对肿瘤细胞DNA损伤修复基因表达的影响。方法用青蒿琥酯处理K562细胞72 h,倒置光显微镜下观察细胞形态变化,Trizol法提取总RNA,RT-PCR检测DNA依赖性蛋白激酶(DNA-PK)的表达变化。结果倒置光显微镜观察到青蒿琥酯处理的K562细胞出现不同程度的皱缩,核分裂相减少,细胞密度下降,漂浮细胞增多。RT-PCR检测结果显示DNA-PK表达下调。结论青蒿琥酯可通过下调K562细胞DNA-PK表达,诱导K562细胞凋亡发挥其抗癌作用。 Objective To investigate the effect of artesunate on expression of damage repair gene in tumor cells by RT-PCR. Methods K562 cells were treated with artesunate for 72 hours, and then cell morphological changes were observed under invelled microscope, total RNA samples were obtained by Trizol and were reversely transcribed to the cDNA, RT-PCR was used to detect the expression changes of DNA-PK. Results The nunmbers of drift cells increased, the denisity of cells decreased, different degrees of shinks occured and nuclear division phases decreased under invert microscope after K562 cells were treated with artesunate for 72 hours. The results of RT-PCR showed down-regulation of DNA-PK. Conclusion Artesunate exerts anticancer effect by downregulating the gene expression of DNA-PK involved in apoptosis.
