小窝蛋白-1在CTD-ILD肺组织中的表达及TGF-β1对其表达调控的体外研究 被引量:3

摘要 目的:了解小窝蛋白-1(Caveolin-1)在结缔组织病相关肺间质病变(CTD-ILD)患者肺组织中的表达,及转化生长因子-β1(TGF-β1)对小窝蛋白-1在人肺成纤维细胞(HLF)中表达的影响。方法:CTD-ILD患者及正常对照肺组织冰冻切片行小窝蛋白-1免疫荧光染色;原代人肺成纤维细胞培养、鉴定,选用11~20代HLF,以终浓度分别为0、1、3、5、10、15μg/L的TGF-β1诱导24h以及浓度为10μg/L TGF-β1分别诱导0、6、12、24、48h,用定量PCR及Western Blot方法检测小窝蛋白-1mRNA及蛋白表达变化,实验重复3次。并且于HLF中加入10μg/L TGF-β1诱导24h,观察诱导前后小窝蛋白-1的荧光表达变化。结果:小窝蛋白-1在CTD-ILD患者肺间质中的表达与对照组相比减弱;小窝蛋白-1主要表达于人肺成纤维细胞的胞膜上,经TGF-β1诱导可使其表达减弱,且呈现一定的浓度和时间依赖趋势。与TGF-β1 0μg/L(对照组)相比,小窝蛋白-1mRNA表达在5、10、15μg/L TGF-β1组均下降,差异均有统计学意义(P<0.01);而小窝蛋白-1的蛋白表达仅在10、15μg/LTGF-β1组有减少,差异有统计学意义(P<0.01)。以TGF-β1作用0h组为对照,小窝蛋白-1mRNA表达在TGF-β1作用6h、12h、24h、48h组均有下降,差异均有统计学意义(P<0.01);而小窝蛋白-1蛋白表达则在TGF-β1作用24h、48h组下降,差异有统计学意义(P<0.01)。结论:小窝蛋白-1在CTD-ILD患者肺组织中的表达减少,TGF-β1可诱导小窝蛋白-1表达下降,小窝蛋白-1的下调可能参与了CTD-ILD的发生和发展。
