补肾益精法对硬皮病成纤维细胞SMAD3/FLI1平衡的影响

Research on Bushen Yijing Therapy to the Balance of SMAD3/FLI1 in Fibroblast of Scleroderma

目的:初步研究补肾益精法中药复方对硬皮病成纤维细胞SMAD3/FLI1"平衡"的影响。方法:体外培养建立硬皮病组和正常对照组皮肤成纤维细胞系;使用补肾益精法中药复方含药血清与对照血清处理两组细胞;采用实时定量RT-PCR技术测定I型前胶原COLⅠ-α2与TGF-β途径中SMAD3、FLI1等因子的基因转录水平。结果:补肾益精法可不同程度地减少硬皮病成纤维细胞COLⅠ-α2、SMAD3表达,上调FLI1基因表达,使SMAD3/FLI1比值趋向健康对照组水平。结论:补肾益精法对硬皮病成纤维细胞表型的影响可能与TGF-β途径中SMAD3/FLI1"平衡"有关。

Objective： To study the effect of Bushen Yijing Therapy on the SMAD3/FLI1 balance in the scleroderma fibroblasts. Methods： The scleroderma and normal fibroblast cell lines were established in vitro. The fibroblast cell lines were man- aged with Bushen Yijing Therapy or control therapy. The transcriptions of COL I -0.2, SMAD3 and FLI1 were detected by realtime quantitative RT-PCR respectively. Results： To some extent, Bushen Yijing Therapy could decrease the transcriptions of COL I -a2and SMAD3, could increase the expression of FLI1 and normalize the SMAD3/FLI1 ratio. Conclusion： The influence of the scleroderma fibroblast phenotype by Bushen Yijing therapy may be related to the SMAD3/FLII ＂balance＂ in the TGF-β pathway.
