经静脉移植骨髓基质干细胞治疗脊髓损伤后碱性成纤维生长因子表达变化

摘要 目的:通过观察骨髓基质干细胞(BMSCs)移植治疗脊髓损伤后对bFGF表达的影响,探讨经静脉移植骨髓基质干细胞治疗脊髓损伤的可行性。方法:用改良的Allen’s WD法建立脊髓(T12节段)损伤模型,免疫组织化学方法检测bFGF的表达。结果:bFGF在各组大鼠脊髓中均呈阳性表达,损伤后对照组与BMSCs移植组各个时点的表达量仍高于正常组(P<0.05),7d最高,后渐下降,但21d表达仍高于正常。其中BMSCs移植组与对照组各个时点的bFGF表达量均有显著差异(P<0.05)。结论:BMSCs移植治疗大鼠脊髓损伤模型有助于脊髓功能的恢复。 Objective:To explore the feasibility of treating spinal cord injury by transplantation of bone marrow stromal cells via vein through watching the change of basic fibroblast growth factor (bFGF). Methods: Spinal cord injury (T12 segment)model was established by modified Allen's WD method. The expression of bFGF was detected with immunohistochemical method.Results: bFGF was expressed in the rat spinal cord of all three groups.After the injury,the expression level of bFGF at each time point in both spinal cord injury and BMSCs groups was significantly higher than that in normal groups (P〈0.05) ,reached a peak at 7d,then decreased gradually,but still higher than normal level at 21d.The bFGF expression level was significantly different between BMSCs group and control group (P〈O.05)at each time point.Conclusion: These results suggest that BMSCs transplantation is helpful to promote the functional recovery in spinal cord injury model.
