摘要
目的:建立HPLC法测定火绒草中木犀草苷和咖啡酸的含量方法。方法:采用高效液相色谱法,色谱柱:Agilent TC-C18色谱柱(250 mm×4.6 mm,5μm);流动相:乙腈-0.5%冰醋酸(梯度洗脱);流速:1 mL/min;检测波长:咖啡酸的检测波长为320nm,木犀草苷的检测波长为350nm;柱温:25℃。结果:木犀草苷在范围内线性关系良好,r=0.9996;平均回收率为101.35%,RSD=1.58%;咖啡酸在范围内线性关系良好,r=0.9997;平均回收率为98.7%,RSD=2.34%。结论:该方法简便、灵敏、准确,可用于火绒草药材的质量控制。
Objective: To establish a HPLC method for the determination of galuteolin and caffeic acid in Leontopodium Leontop odioides ( wild. ) Beauv. Methods: HPLC was used in the content analysis of galuteolin and caffeic acid in the sample. Column : Agilent TC -C18 (250 mm×4. 6 mm, 5μ m), mobile phase: aeetonitrile-0. 5% glacial acetic acid (gradient elution), flow rate: 1 mL/min, wavelength: the wavelength of eaffeie acid is320nm and the wavelength of galuteolin is 350nm, column temperature: 25℃. Results: A good linearity of galuteolin was in the rang of 0. 148μg -0. 74μg and r =0. 9996. The average recovery of galuteolin was 101.35% and RSD = 1.58%. A good linearity of caffeie acid was in the rang of 0. 076μg -0. 38μg and r = 0. 9997. The average recovery of caffeic acid was 98.7% and RSD = 2. 34%. Conclusion: This method is simple, sensitive, accurate, which can be used to control the quality of Leontopodium Leontop odioides ( wild. ) Beauv.
