K-ras与p33ING1、CyclinE在直肠癌组织中表达及生物学关系的研究

摘要 目的探讨K-ras蛋白与p33ING1蛋白、CyclinE在直肠癌组织中表达情况及其与直肠癌生物学行为的关系。方法运用S-P免疫组化技术,检测72例直肠癌组织及正常直肠黏膜组织中K-ras蛋白与p33ING1蛋白、CyclinE的表达。结果 K-ras与p33ING1、CyclinE的蛋白表达阳性率在直肠癌组织中分别为62.5%、29.2%、72.2%,在正常直肠黏膜中分别为2.8%、91.7%、18.1%,差异有统计学意义(P<0.05);K-ras蛋白在直肠癌组织中的表达与年龄、肿瘤大小、临床分型、组织分化无关;与淋巴结转移及Dukes分期有关,不同分组间的K-ras蛋白表达率差异有统计学意义(P均<0.05);K-ras蛋白表达和p33ING1蛋白表达呈负相关(rs=-0.404,P<0.05),K-ras蛋白表达和CyclinE表达呈正相关(rs=0.571,P<0.05)。结论直肠癌组织中K-ras蛋白、CyclinE表达增高,抑癌基因蛋白p33ING1表达降低,提示K-ras与p33ING1、CyclinE参与直肠癌的发生、发展,并在直肠癌形成过程中起着重要作用。并可能为直肠癌的早期诊断和治疗提供新的方法。 Objective To explore the expression and biological behaviors relation of K-ras protein and p33ING1 proteins, CyclinE in rectal cancer tissues. Methods S-P immunochemistry technique was applied to detect the expression of K-ras protein and p33ING1 proteins, CyclinE in 72 cases of normal rectal mucosa and rectal cancer tissues. Results The positive expression rates of K-ras protein and p33ING1 proteins, CyclinE were 62. 5%, 29. 2% ,72. 2% in rectal cancer tissues, and 2. 8% ,91.7%, 18. 1% in normal mucosa. The first group performed significant difference with the second group( P 〈 0. 05). There was a negative correlation between the expression of K- ras and the expression of p33ING1 ( r~ = - 0. 404, P 〈 0. 05 ). K-ras protein expression rate increased in the organizatioris which were poorly differentiation, had metastatic lymph nodes and high clinical stage, the difference was statistically significant( P 〈 0. 05 ). The expression of K-ras protein was positively related with the expression of CyclinE(r8 = 0. 571 ,P 〈 0. 05 ). Conclusions In rectal cancer tissues, the expression of K-ras protein ascend and the expression of p33ING1 protein, CyclinE descend. K-ras and p33ING1, CyclinE may be participate in rectal cancer occurrence, development, and play an important role in the process of formation of rectal cancer. This maybe provides a new method for early diagnosis and treatment of rectal cancer.
