摘要 目的本研究旨在研究金属基质蛋白酶(MMPs)在小鼠药理性孕酮撤退月经模型中的定位。方法卵巢切除的小鼠在诱导蜕膜化之后给予米非司酮处理,免疫组化检测金属基质蛋白酶MMP3和MMP7定位。结果 MMP3从米非司酮处理后0 h到24 h,存在于腔上皮下的局部基质、上皮、蜕膜化区域以及基底层和功能层的交界处;从32 h到48 h存在于腔上皮以及其下基质区域。MMP7从0 h到24 h存在于基底层,上皮和中间蜕膜化区域,从32 h到48 h与MMP3的定位相一致。结论 MMP3和MMP7蛋白质在此模型中定位呈区域性分布。 Objective: To investigate the expression of matrix metalloproteinases (MMPs) in a mouse menstrual- like model created by pharmacologic progesterone withdrawal. Methods: The menstrual-like model was prepared by administration of mifepristone after decidualization of ovariectomized mice. Immunohistochemistry was performed to detect the locations of MMPs proteins in the uterus. Results: MMP3 was present in the focal subluminal stromal zone, epithelium, decidual zone and interface between the basal zone and functional zone from 0 h to 24 h after administration of mifepristone, and epithelium and subluminal epithelium stroma from 32 h to 48 h. MMP7 was localized in the basal zone, epithelium and mid decidual zone from 0 h to 24 h, and a similar localization to MMP3 from 32 h to 48 h.Conclusions. The localization of MMP3 and MMP7 proteins in the model displayed a regional characterand functioned in endometrium repair, which was consistent with those previously reported data in the mouse model by physiological progesterone withdrawal and in primates.
