摘要
目的研究静压力对体外人牙周膜成纤维细胞(human periodontal ligament fibroblasts,HPDLFs)增殖活性的影响。方法用静压力加载装置对第4代HPDLFs分别施加0、15、30、45 kPa的持续性静压力,加力时间为1 h。加力后采用流式细胞术及噻唑蓝(methyl thiazolyl tetrazolium,MTT)比色法观察HPDLFs增殖活性。结果流式细胞术检测结果显示0、15、30 kPa静压力下HPDLFs增殖指数分别为(40.11±0.59)%、(49.89±0.75)%、(55.77±1.57)%,呈递增趋势(P<0.05);静压力达到45 kPa时,HPDLFs增殖指数有所降低,为(35.35±0.85)%。MTT法检测结果显示在0、15、30 kPa静压力下吸光度值分别为0.30±0.04、0.35±0.05、0.40±0.07,呈递增趋势(P<0.05);静压力达到45 kPa时,吸光度值有所降低,为0.25±0.04。结论适当大小的静压力能促进HPDLFs的增殖,过大的静压力会抑制HPDLFs的增殖。
Objective To analyze the effects of different static compressive stress on the proliferation of human periodontal ligament fibroblasts(HPDLFs) in vitro.Methods The cultured HPDLFs of the fourth passage were subjected to the different static compressive stress with a magnitude of 0,15,30,45 kPa,respectively,for 1 hour.Then the proliferation was detected by flow cytometry and methyl thiazolyl tetrazolium(MTT) assay.Results Under the compression with a magnitude of 15,30 kPa,the proliferation of HPDLFs greatly increased(P〈0.05),but dropped under the compression with a magnitude of 45 kPa.Conclusion Compression at a moderate magnitude could enhance the proliferation,and too excessive compression could produce negative effects on the normal metabolism.
