甘草β-AS基因时空表达模式研究 被引量:5

摘要 目的:揭示甘草β-AS基因表达的时间特异性及空间特异性。方法:通过PCR扩增获得不同时间、不同部位甘草β-AS基因的cDNA序列,琼脂糖凝胶电泳后用Gel-Pro分析软件对电泳条带进行光密度分析,计算其相对表达量。结果:空间特异性实验显示β-AS基因在甘草的地上部分没有表达,地下组织中,新陈代谢最旺盛的根尖部分表达量高于根茎。时间特异性实验显示甘草β-AS基因在全年中的表达水平可分为4个阶段。12月至2月,β-AS基因表达低于检测水平;3月至5月,β-AS基因开始表达,表达量逐渐上升;5、6及8、9月,β-AS基因表达量保持较高水平;10、11月表达量开始下降。结论:在研究甘草β-AS基因时,应该选取转录水平较高的5、6及8、9月份;选取转录水平较高的根尖作为提取总RNA的部位。 Objective:To reveal the temporal and spatial specificity of the expression of β-AS gene of Glycyrriza uralensis. Meth- ods:Used PCR to obtain the cDNA of β-AS gene of Glycyrriza uralensis at different time and from different part and Gel-Pro to carry out the densitometric analysis of the electrophoretic band, then calculate the relative expression. Results : The spatial specificity experiment showed that β-AS gene didn't express in the overground part of Glycyrriza uralensis ,while in the underground part, the expression of β- AS in root tip was higher than that of rootstock. And the temporal specificity experiment showed that the expression of,β-AS gene of β- cyrriza uralensis could be divided into 4 stages. From December to February ,the expression of β-AS gene was under the detection limit. From March to May,β-AS gene began to express. From May to September,the expression of β-AS gene kept at a high level. And in Oc-tober and November the expression of β-AS gene began to decrease. Conclusion: When the β-AS gene of Glycyrriza uralensis is re- searched, the root tip is the suitable plant material and May ,June, August and September are the right acquisition time.
