炎琥宁注射液在细胞水平上抑制CVA16病毒的初步评价

目的通过体外实验,观察利注射液炎琥宁注射液对肠道病毒C型病毒在横纹肌肉瘤传代细胞(RD-A细胞)复制的抑制作用和体外灭活作用。方法使用从病人咽拭子样本中分离的病毒,通过三种体外细胞实验:药物在细胞中抑制病毒CVA16复制的药效测定实验;药物对细胞的保护作用实验;药物体外对CVA16病毒的灭活作用的药效测定实验。观察炎琥宁注射液对CVA16病毒感染横纹肌肉瘤传代细胞(RD.A细胞)中所起的作用。结果在炎琥宁注射液在细胞中抑制CVA16病毒复制的药效测定实验中1,:640炎琥宁注射液稀释度组较病毒对照组延迟病变出现2 d,细胞生长未受到影响。在药物对细胞的保护作用实验中l,:8炎琥宁稀释度组较病毒对照组延迟病变出现4 d。在炎琥宁注射液药物体外对CVA16病毒的灭活作用的药效测定研究中1,:40稀释度组较病毒对照组延迟病变出现2 d,而细胞生长未受到影响。结论炎琥宁注射液在体外有抑制CVA16病毒复制和部分灭活病毒的作用;并有一定的保护细胞抑制CVA16病毒感染的作用。本研究用于临床上CVA16感染的早期预防及治疗具有一定的指导意义。

Objective Observe the effect of Potassium Sodium Dehydroandroan drographolide Succinate for Injection on reducing the CVA16 replication, inactivating CVA16 and protecting the RD - A cells against the CAV16 infection in vitro. Methods Using the CVA16 isolated from throat swab of patient, the effect of Potassium Sodium Dehydroandroan drogra- pholide Succinate for Injection on RD - A cells during the CVA16 infection was observed. Results In the experiment of Potassium Sodium Dehydroandroan drographolide Succinate inhibiting the CVA16 replication, comparing with the negative control group, the group of 1：640 dilution of Potassium Sodium Dehydroandroan drographolide Succinate delayed the CPE for 2 days ,while the normal cell group was growing very well. In the experiment of protecting cell from CVA16 infection. The group of 1：8 dilution of Potassium Sodium Dehydroandroan drographolide Succinate delayed the CPE for 4 days. In the ex- periment of its effect on the inactivation of CVA16o the group of 1：40 dilution of Potassium Sodium Dehydroandroan drogra- pholide Suecinate delayed the CPE for 2 days comparing with the virus control group. Conclusion Potassium Sodium De-hydroandroan drographolide Suceinate could inhibit the repheation of the CVA16 and inactivate the CVA16 in vitro. Addi- tionally. Potassium Sodium Dehydroandroan drographolide Suceinate could protect RD - A cells from CVA16 infection. The observation will contribute to CVA16 infection control and quick medicine therapy.
