摘要
目的采用蒸发光散射检测器(ELSD)测定麦冬中麦冬皂苷D、D'的含量。方法 Waters sy-meteryC18色谱柱(4.6 mm×150 mm,5μm),水-四氢呋喃-乙腈(52∶3∶45)等度洗脱,流速1.0 mL·min-1,柱温30℃,增益值10,压力2.5 Pa,飘移管温度65℃。结果麦冬皂苷D和麦冬皂苷D'分别在48.60~972μg·mL-1,24.75~495μg·mL-1范围内具有良好的线性关系,平均回收率(n=6)分别为101.7%(RSD=2.69%)和97.7%(RSD=3.25%)。结论该方法快速、准确,可用于麦冬药材中麦冬皂苷D和麦冬皂苷D'的含量测定。
Objective To determinate of ophiopogonin D and ophiopogonin D' from Ophiopogon japonicus by ELSD-HPLC. Methods ELSD-HPLC conditions were as the follows:Waters Symmetry Cls column (4.6 mm × 250 mm, 5μm) by gradient elution with water, tetrahydrofuran and acetonitrile (52:3:45) , the flow rate 1.0 mL ·min-1 , the column temperature maintained at 30 ℃, the gain 10, the pressure 2.5 pa, and the draft temperature 65 ℃. Results The calibration curve showed a good linearity with the range of 48.60 - 972 μg mL- 1 for ophio- pogonin D and 24.75 - 495μg mL-1 for ophiopogonin D', respectively. The recovery (n = 6) was 101.7% (RSD = 2.69% ) and 97.7% (RSD = 3.25% ), respectively. Conclusion This method was simple and accurate and could be used for the determination of ophiopogonin D and ophiopogonin D' from Ophiopogon japonicus.
