摘要
目的建立高效液相色谱法测定还阳参药材中绿原酸的方法。方法采用Sino Chrom DDS-BP色谱柱(250mm×4.6mm,5μm),流动相为乙腈-0.4%磷酸水溶液(13:87),体积流量1.0mL/min,检测波长327nm,柱温25℃。结果绿原酸进样量在0.046~0.644μg与峰面积呈良好的线性关系(r=1),样品平均加样回收率为101.9%,RSD值为1.22%(n=6);检出限和定量限分别为0.01ng和0.04ng。结论此方法简便、准确、重复性好,可用于还阳参药材的质量控制。
Objective To establish a method of HPLC for the determination ofchlorogenic acid in the herbs of Crepis turczaninowii. Methods The samples were separated on a Sino Chrom DDS-BP column (250 mm x 4.6 mm, 5μm) with the mobile phase of acetonitrile-0.4% phosphoric acid (13:87). The flow rate was 1.0 mL/min; The detection wavelength was set at 327 nm; And column temperature was 25 ℃. Results There was a good linear relationship between the chlorogenic acid and peak area in the range of 0.046--0.644μg (r = 1). The average recovery rate was 101.9%, and RSD was 1.22% (n = 6). Conclusion The method is simple, accurate, and highly reproductive, and could be used for the quality control of the herbs of C turczaninowii.
