摘要
目的考察水提醇沉法和乙醇煎煮回流法对蟾皮有效成分的影响,优选蟾皮提取工艺。方法通过正交试验法,以华蟾酥毒基为指标,采用高效液相色谱法对不同方法提取的蟾皮样品进行含量测定。结果水提醇沉法各因素影响大小为提取次数〉加水倍数〉提取时间,最佳工艺为B3A3C2,即加10倍量的水,煎煮3次,每次煎煮时间为45min。乙醇煎煮回流法各因素影响大小为乙醇浓度〉提取时间〉加醇倍数,最佳工艺为A1B3C3,即加30倍量的50%乙醇,煎煮3次,每次煎煮时间为1h。结论综合评分结果显示乙醇煎煮回流法优于水提醇沉法,但是水提醇沉法提取的华蟾素毒基含量更高。
Objective To optimize the extraction technology by comparing the influence of water extracting-alcohol precipitating with ethanol decoction reflux methods on active composition in the skin of bufo bufo gargarizans cantor. Methods HPLC method was employed to determine the content of Cinobufagin, the skin of Bufo bufo gargarizans cantor samples extracted with different solvents and methods. Orthogonal experiment design was employed to screen the optimal conditions. Results The result of water extracting-alcohol precipitating method demonstrated that the influence order of factors was following: extraction times; adding water multiples; and extraction time. The optimum technology is B3A3C2 of which was adding i0 times amount of water, extracting 3 times and every time with 45 minutes. Ethanol decoction reflux method results showed that the influence order of factors was ethanol concentration 〉 extraction time 〉 add alcohol ratio, and the optimum technology is A1BsC3 of which was adding 30 times the amount of 50% ethanol, extracting 3 times and every time with 1 hour. Conclusion Composite score results reflected that ethanol decoction reflux was better than ,'rater extracting-alcohol precipitating method. According to the content of Cinobufagin, however, water extracting-alcohol precipitating method was more effective.
