摘要
目的:建立磐安产延胡索的HPLC指纹图谱鉴别方法,比较遗传变异的大叶型和小叶型延胡索之间的区别。方法:以延胡索乙素为参照物,采用HPLC检测方法:色谱条件:Ultimate LP-C18色谱柱(4.6mm×250mm,5μm),乙腈-三乙胺乙酸溶液(每1000mL水中加冰乙酸30mL、三乙胺8mL)二元梯度洗脱,体积流量0.8mL/min,检测波长280nm,进样量20μL,柱温35℃。结果:确定了11个共有峰,建立了延胡索的HPLC指纹图谱,比较研究了大、小叶型延胡索不同加工方法的样品,结果相似度均在0.90以上。结论:本法操作简便,重现性好,可有效控制延胡索的质量,并为产地优质品种选育提纯及加工提供实验依据。
Objective: To establish the HPLC fingerprint identification method of Rhizoma Corydalis produced in Pan'an and to compare the differences between large leaf type and lobular type of Rhizoma Corydalis by genetic variation. Methods: The method was determined with tetrahydropalmatine as the reference by HPLC, chromatographic conditions were performed on Uhimate LP - C ts column (4.6mm×250mm,5μm), with acetonitrile - triethylamine acetic acid ( per 1000mL water plus acetic acid 30mL, triethylamine 8mL)as gradient elution. The detection wavelength was 280nto with the volume flow rate of 0.8mL/min,injection volume was 20μL,the column temperature was 35℃. Results: The method was identified 11 peaks, the HPLC fingerprint of Rhizoma Corydalis was established. The similarity of results were above 0.90, compared the large leaf and lobular type of Rhizoma Corydalis samples from different processing methods. Conclusion : The method is simple and has good reproducibility, it can be used to effectively control the quality of Rhizoma Corydalis, and provide experimental evidence for purifying and processing varieties.
