摘要
目的:探讨MECP2基因多态性与海洛因依赖的关联性。方法:在32名健康对照者中对MECP2基因进行测序分析,从测序结果中选择4个SNP位点在331名海洛因依赖者和325名健康对照者中用Snapshot技术进行基因分型;根据调查量表信息将病例组中的男性患者分为高低剂量两组;用HaploView 4.2及SPSS 15.0软件分析SNP与药物依赖的相关性。结果:海洛因依赖组与对照组MECP2基因rs2075597、rs61751363和rs189253298的基因型及等位基因频率分布比较,差异无统计学意义(P>0.05)。rs189253298基因型频率在低剂量组和高剂量组中的分布比较,差异有统计学意义(P=0.012)。结论:MECP2基因的rs189253298多态性位点与海洛因依赖者成瘾后每天海洛因的使用量相关,携带有G等位基因的个体每天使用的海洛因量低,是一个保护因素。
ABSTRACT Objective:To explore the association between methyl- CpG- binding protein 2(MECP2) gene polymorphisms and heroin addiction. Methods:MECP2 gene was sequenced in 32 healthy controls to identify the polymorphisms. Four SNPs in MECP2 were genotyped in 331 heroin addicts and 325 healthy controls using the method of Snapshot. According to average drug use dosage per day, male heroin addicts were divided into two subgroups: low dosage group (≤ 0.4 g)and high dosage group (≥ 0. 5 g). HaploView 4.2 and SPSS 15.0 software were used to perform the association analysis between these polymorphisms and heroin addiction. Results:No significant difference in 3 SNPs allele and genotype frequencies were observed between heroin addicts and controls. The G allele frequency of rs189253298 was significantly higher in low dosage group compared with high dosage group (P = 0. 012). Conclusion:The polymorphism rs189253298 of MECP2 was associated with heroin dosage in drug addicts.
