摘要
目的:研究mdr1b/a基因siRNA(small interfering RNA)逆转膀胱癌的多药耐药性。方法:设计mdr1b/a基因的siRNA,转染膀胱癌T24/ADM细胞,用RT-PCR分析mRNA的表达,Rh123分析P-gp活性,流式细胞仪检测细胞内阿霉素积累量。结果:siRNA能明显地逆转T24/ADM细胞的多药耐药,使mdr1b/a基因的mRNA表达水平下调,细胞内阿霉素积累量显著增加(P<0.05)。结论:mdr1b/a基因siRNA能够逆转膀胱癌的多药耐药性。
Objective: To investigate the effect of mdrlb/mdrla siRNA on multidrug resistance of bladder carcinoma cell line T24/ADM. Methods: The proper mdrlb/a siRNA was designed, synthesized and transfected into bladder carcinoma cell line T24/ADM. Expression level of mdrlb/a mRNA was deter- mined by PCR. The activity of Pgp was measured by Rhodamine 123 efflux assay, The intracellular accumulation of ADM was assayed with flow cytometry. Results: After treated with siRNA, the expression level of mdrlb/ was increased si ante of bladder a mRNA in T24/ADM decreased obviously, and the intracellular accumulation of ADM gnificantly (P〈0.05). Conclusions: The mdrlb/asiRNA can reverse the multidrug resist- carcinoma cell line T24/ADM.
