摘要
目的观察不同浓度银杏叶提取物对缺氧状态下视网膜血管内皮细胞增殖的影响。方法取第7代牛视网膜血管内皮细胞用于实验,采用二氯化钴(CoCl2)建立细胞化学缺氧模型,四甲基偶氮唑盐(MTT)比色法检测血管内皮细胞的增殖情况。结果在CoCl2所诱导的化学缺氧下,视网膜血管内皮细胞增殖减少。与缺氧组比较,0.001、0.01、0.1μg/ml银杏叶提取物组作用24 h及48 h后,血管内皮细胞光密度值均增加(P<0.05)。结论银杏叶提取物对缺氧状态下的视网膜血管内皮细胞有明显的促进增殖作用,其作用与银杏叶提取物的剂量有关。
OBJECTIVE To investigate the proliferation effect of Ginkgo Biloba Extract (EGB)on retina vascular endothelial cells ( RVECs ) induced by hypoxi. METHODS The seventh generation of RVECs was cultured for the research. COCl2 was used to set up the cell chemistry hypoxia mode. The effect of Ginkgo Biloba Extract on the proliferation of cells was measured by methyl thiazolyl thiazolium (MTr) assay. RESULTS COC12 could decrease the proliferation of RVECs. Compared with hypoxia group, RVECs cultured with 0.001,0.01 and 0.1μg/ml Ginkgo Biloba Extract showed significant proliferation effect at 24 h and 48 h (P〈0.05). CONCLUSIONS Ginkgo Biloba Extract had obvious proliferation effect on RVECs induced by hypoxi. The effect was related to the Ginkgo Biloba Extract' s dose.
