摘要
目的探究布鲁氏菌减毒活疫苗株与亲本株的分子遗传差异。方法通过布鲁氏菌全基因组DNA芯片对4株减毒活疫苗株进行比较基因组学研究。结果通过比较基因组学研究,发现疫苗菌株中存在大片段基因缺失,同时发现一些基因以多拷贝形式存在。4株减毒活疫苗株分别缺失不同数目的功能基因,主要为胞内活动有关基因和功能未知基因(分别为31和45个)。较深入地认识了布鲁氏菌减毒活疫苗株基因构成上的差异。结论与亲本株相比,4株减毒活疫苗株缺失大量基因,构成了不同疫苗株差异的遗传基础。
Four live attenuated vaccine strains were analyzed by comparative genomic hybridization to explore the molecular genetic variation between live attenuated Brucella vaccines and their parental strains.Results demonstrated that there were a number of deletion fragment regions(DFRs) and duplications genes in these vaccine strains.A number of functional genes were missed in these four live attenuated vaccine strains,which were mainly concerned with intracellular processes and unknown function(31 and 45 genes,respectively).The differences in genes content of four vaccine strains implied their possible mechanism of attenuation,immunogenicity and protection.Compared with the parental strain,a large number of missing genes constitute the genetic variations in these four vaccine strains.
