摘要
目的观察原花青素(PC)对瘦素诱导的肝星状细胞(HSC)增殖及组织抑制基质金属蛋白酶-1(TIMP-1)产生的影响。方法磺基罗丹明B法检测HSC增殖,利用活性氧(reactive oxygen species,ROS)探针2,7-二氯二氢荧光素二乙酯检测ROS,酶联免疫吸附实验测定TIMP-1的浓度。结果 PC在50、100、200μg/mL时能够抑制瘦素诱导的HSC的增殖(P<0.05),同时能够抑制瘦素诱导的ROS的产生(P<0.05),上述PC各浓度组亦能够抑制TIMP-1的产生(P<0.05),浓度之间抑制效果具有统计学差异(P<0.05)。结论 PC可能通过抑制瘦素诱导的氧化应激,抑制HSC的增殖以及TIMP-1的产生。
Objective To observe the impacts of procyanidins (PC) on the proliferation of hepatic stellate cell (HSC) and production of tissue inhibitor of matrix metalloprotease-1 (TIMP-1) induced by leptin. Methods Sulforhedamine B assay was performed to test the proliferation of HSC. The probe of reactive oxygen species (ROS) 2, 7-dichlorfluo- rescein-diacetate was used to detect the production of ROS. Enzyme-linked immunosorbent assay was used to detect the concentration of TIMP-1. Results PC at concentrations of 50, 100,200 ixg/mL could inhibit the proliferation of HSC induced by leptin (P 〈0.05). Meanwhile, it had the inhibitory effect on the production of ROS (P 〈0.05). The PC groups above could also inhibit the generation of TIMP-1 ( P 〈 0.05 ). There were statistically significant differences among the groups (P 〈0.05). Condusion PC might inhibit the HSC proliferation and TIMP-1 production through inhibiting the oxidative stress induced by leptin.
