摘要
目的探讨尼莫地平对H2O2引起的血管内皮细胞凋亡的影响。方法以体外培养的人脐静脉内皮细胞为研究对象,通过TUNEL法、流式细胞术来检测H2O2对血管内皮细胞凋亡的影响以及尼莫地平的干预作用。结果 H2O2(100μmoI/L、200μmol/L、400μmol/L)明显增加内皮细胞的凋亡率,而尼莫地平(1.2μg/ml)能拮抗H2O2(200μmol/L)引起内皮细胞的凋亡作用。结论尼莫地平对H2O2引起的血管内皮细胞凋亡有拮抗作用。
Objective To study the effect to Nimodipine on vascular endothelial cell(ECV-304) apoptosis induced by various concentrations of H2O2. Methods Cultured vascular endothelial cells were treated with various concentrations of H2O2 and Nimodipine for 24 hours. The apoptosis rate of vascular endothelial cells was anaylzed with TUNEL and flow cytometer. Results H2O2 was able to induce apoptosis in human vascular endothehal cells. Nimodipine could inhibit apoptosis of them. Conclusion Nimodipine could protect vascular endothelial cells from various concentrations of H2O2 induced apoptosis.
