玉竹生物活性成分C对小鼠免疫功能的影响 被引量:6

摘要 研究玉竹生物活性成分C对小鼠细胞免疫功能的影响,并探讨其作用机制。采用MTT比色法和CFSE检测小鼠淋巴细胞转化情况,流式细胞术检测对αβT和γδT细胞增殖及CD4+αβT细胞/CD8+αβT细胞比值的影响,ELISA法检测脾细胞培养上清中Th1细胞因子IFN-γ和Th2细胞因子IL-4的分泌水平。结果显示,玉竹生物活性成分C在一定浓度范围内可促进小鼠脾αβT细胞增殖,其中以1 000μg/mL的浓度作用最强。1 000μg/mL浓度的玉竹生物活性成分C具有降低CD4+αβT细胞与CD8+αβT细胞比值,提高CD8+αβT细胞数量的作用。1 000μg/mL浓度的玉竹生物活性成分C能有效刺激小鼠脾淋巴细胞释放细胞因子IFN-γ,而对IL-4的产生没有明显的影响。本研究为开发玉竹生物活性成分C提供实验依据。 Effects of bio-active component C from yuzhu ( a kind of TCM ) ( Polygonatum odoratum) ( YZBCC ) on mice immune function were studied and investigated its effective mechanism. MTT colorimetry and CFSE were adopted to detect the condition of lymphocyte transformation in mice, the proliferation of αβT and TST cells and the ratio of CD4 αβT cell/CD8 αβT cell were detected by flow cytometry and the secretion level of Thl cytokincs IFN-T and Th2 cytokine IL-4 in spleen cell culture supernatant were detected by ELISA. The results showed that YZBCC could promote the proliferation of spleen αβT cells at a certain concentration range, and the strongest one was at 1 000 μg/ mL. At the concentration of 1 000 μg/mL POBCC had the effect to reduce the ratio of CD4αβT cell and to increase the number of CD8 + αβT cell. At the concentration of 1 000 μg/mL YZBCC could effectively stimulate spleen lym- phocytes of mice to release cytokines IFN-γ, however, un-noticeable effect on the production of IL-4. This study pro- vides experimental bases to develop the YZBCC.
