摘要
目的 研究血小板激活因子 (PAF)在急性脑缺血过程中的作用。方法 用高效薄层层析法对大鼠全脑缺血后再灌注 1、3、6、72小时血液、海马、大脑顶叶皮质的 PAF进行了观察。结果 再灌注 1小时后海马中 PAF含量较对照组明显升高 (P<0 .0 1) ,而 3小时时则下降至正常水平 (P>0 .0 5 )。血液中 PAF的升高出现较晚 ,再灌注后 3小时时血中 PAF明显升高 ,6小时时最明显 ,72小时时与对照组比较仍有显著性差异(P<0 .0 5 )。结论 脑缺血后海马 PAF的变化快而短暂 ,早期血液中 PAF可能主要来自脑组织 ,与血脑屏障破坏有关。提示
Objective To investigate the role of platelet activating factor (PAF) in rats after acute cerebral ischemia.Methods After rat global cerebral ischemia and reperfusion for 1,3,6 and 72 h,Wistar rats were killed and the PAF in rats' blood,hippocampus and cerebral parietal lobe cortex was measured with high performance thin layer chromatography.Results after reperfusion for 1 h PAF in hippocampus was significantly higher than the controls( Ρ <0.01),and it decreased to the normal level at 3 h ( P >0.05).The elevation of PAF in blood showed later,after reperfusion for 3 h it was significantly higher than the controls,and it was most remarkable at 6 h.There was remarkable difference between 72 h and the controls( P <0.05).Conclusion The change of hippocampus PAF was quick and short after cerebral ischemia.PAF in blood of early stage might come from brain tissue,and related to the destruction of the blood brain barrier.The study suggested that PAF might play an important role in the pathophysiologic course on early stage of cerebral ischemia.
