钒酸氧化法测定血清总胆红素、直接胆红素的方法学评价 被引量:6

摘要 目的 :WAKO钒酸氧化法与改良J -G法测定胆红素的方法学比较。方法 :采用WAKO钒酸氧化法胆红素测定试剂盒同时测定血清中的总胆红素、直接胆红素 ,与改良J -G法测定胆红素的方法进行了比较 ,并对钒酸氧化法的重复性试验、线性试验、干扰试验、回收试验进行了评价。结果 :钒酸氧化法(Y)与改良J-G法 (X)有很好的相关性 (总胆红素 :Y =0 992 6X + 2 2 344,r =0 9991 ;直接胆红素 :Y =0 9968X + 1 1 694,r=0 9982 ) ;测定总胆红素的线性可达 684μmol/L ,直接胆红素的线性可达342 μmol/L ;测定总胆红素天内重复性变异平均为 1 1 7% ,天间重复性变异平均为 1 88% ,直接胆红素天内重复性变异平均为 1 88% ,天间重复性变异平均为 2 76% ;溶血、脂浊、维生素C对测定表现为较微的干扰。总胆红素的回收率平均为 98 7% ,直接胆红素的回收率平均为 1 0 2 0 %。结论 :钒酸氧化法是一准确、灵敏的、适用于实际工作的测定胆红素的方法。 Objective Method comparison of Wako vanadate oxidation with improved J-G method for determination of bilirubin.Methods We use the Wako vanadate oxidation reagent of bilirubin to determinate serum total bilirubin and direct bilirubin simultaneously,take a comparison with the method of improved J-G and also take run-in-run test,linear test,recovery test,interfere test of vanadate oxidation.Results Vanadate oxidation( Y )has good correlation with improved J-G (total bilirubin Y =0.992 6 X +2.234 4, r =0.999 1;direct bilirubin Y =0.996 8 X +1 169 4, r =0.998 2).Linear range of total bilirubin can attach to 684 μmol/L and direct bilirubin can attach to 342 μmol/L.The inter day CV for total bilirubin is 1.17% and direct bilirubin is 1.88%.The intra day CV for total bilirubin is 1.88% and direct bilirubin is 2.76%.Hemolyte,lipidemia and vitamin C have rarely interfere.The average recovery rate of total bilirubin is 98.7%,direct bilirubin is 102%.Conclusion Vanadate oxidate method is accurate and sensitive and it can be used in the routine determination of bilirubin.
