摘要
目的:观察960抗癌合剂对人肝癌细胞的凋亡作用,揭示其治疗原发性肝癌作用机理。方法:丫啶橙(AO)染色,琼脂糖凝胶电泳,碘化丙锭染色细胞周期分析(PI-FCM)和末端转移酶DNA断裂点原位标记(TUNELFCM)。结果:经960药物血清处理后,细胞皱缩,深染,AO染色,细胞呈致密浓染的黄绿色荧光;DNA电泳见典型“梯形”带;ANA直方图上见典型凋亡峰(0.95%~27.77%),TUNEL标记率64.12%~70.02%。结论:960合剂可诱导人肝癌细胞发生凋亡,这可能是其抗癌作用主要机制之一。
Aim: To investigate the induction of apoptosis by herbal medicine 960 mixture (960) in human hepatoma cells for demonstration of its mechanism of treatment of primary liver cancer. Methods: The drug serum was derived from rates at Ihour after second administration with 8. 0ml/kg of 960. and added to cultured human hepatoma ce11 line SMMC-7721 while the normal rat serum as control. Cell apoptosis was detected by acridine orange (AO) staining, DNA electrophoresis, Cell cycling analysis (PI-FCM) and TdT enzyme mediated deoxynucleotide nick end labeling method (TUNED by flow cytometry. Results: After treatment of the drug-serum, SMMC-7721 cells became small and condensed, and showed condensed and fragment nuclei under the fluorescence microscope by AO staining. There were characteristic DMA 'ladder' on agarose gel electrophoresis, and Sub-G1 peaks (apoptotic peak) in DNA histogram, the positive labeling rates by TUNEL were among 64. 12%-70. 02%,Conclusion: 960 may induce apoptosis in human hepatoma cells, which led the death of the cancer cells.
