肿瘤相关抗原GCF2高水平表达的肝癌细胞株的筛选 被引量:7

摘要 目的筛选出肿瘤相关抗原GCF2高表达的人肝癌细胞株,为进一步探讨GCF2对肝癌发生发展的可能作用奠定基础。方法选择4种人肝癌细胞株(BEL-7404、HepG2、SMMC-7721和QGY-7703)及1种成人正常肝细胞株(HL-7702),培养后分别制作细胞爬片及提取蛋白后,采用免疫细胞染色技术和Western blot的方法筛选出GCF2表达量高的细胞株。结果免疫细胞染色显示,GCF2在5种细胞中的胞核及胞质中均有表达。GCF2蛋白在肝癌细胞BEL-7404、HepG2、SMMC-7721和QGY-7703的表达要强于正常肝细胞HL-7702,其中以BEL-7404的表达量最高。通过Western blot检测,BEL-7404的GCF2相对表达量为0.875±0.134,较其他细胞株高,差异有统计学意义(P<0.05)。结论 BEL-7404细胞中GCF2蛋白水平的相对表达量最高,可以作为下一步探讨GCF2在肝癌发生发展的可能作用的实验材料。
