热休克应答干预对大鼠心肺复苏后神经细胞凋亡的保护作用 被引量:2

摘要 目的研究热休克蛋白70(HSP70)对大鼠心肺复苏后神经细胞凋亡的影响,探索热休克应答对脑复苏的应用前景。方法将46只成年Wistar大鼠随机分为假手术组、模型组和干预组;模型组和干预组制作心搏骤停(SCA)并心肺复苏的动物模型,假手术组不进行模型制作;模型组仅制作SCA动物模型,不进行热休克应答干预;干预组大鼠SCA模型制作后进行热休克干预,即静脉注射HSP70。分别于6、12 h后检测各组大鼠脑海马区神经细胞凋亡数目、Caspase-3活性,对比分析各组大鼠神经细胞凋亡情况的差异,验证HSP70对大鼠心肺复苏后神经细胞凋亡的保护作用。结果与模型组比较,干预组在6、12 h两时点凋亡细胞数和Caspase-3阳性细胞数明显减少,两组比较差异均有统计学意义(P<0.05)。结论热休克应答干预对大鼠心肺复苏后神经细胞凋亡有保护作用,对脑复苏具一定的治疗前景。
