摘要
目的:研究CW7213对自发性2型糖尿病KKAy小鼠的治疗作用。方法:以胰岛素为阳性对照,给自发性2型糖尿病KKAy小鼠皮下注射CW7213(20、60、120μg.kg-1),每天给药1次,连续4周,分别于首次给药后0、1、2、3 h及1、2、3、4周取血,测定小鼠的空腹血糖浓度;给药第3周,对小鼠给药15 min后灌胃给予2.5 g.kg-1葡萄糖,负荷0.5、1、2、3 h后取血,测定血糖浓度;末次给药1 h后,测定小鼠空腹血清胰岛素、甘油三酯和总胆固醇浓度。结果:CW7213各剂量组在给药后不同时间段其小鼠空腹血糖浓度与空白对照组相比,呈剂量相关性降低且有显著差异(P<0.01,P<0.05);CW7213连续给药4周后,与空白对照组相比,小鼠的血清胰岛素浓度显著增加,高、中剂量组的甘油三酯浓度下降(P<0.01,P<0.05),对血清总胆固醇未见明显影响。结论:CW7213单次或连续给药对自发性2型糖尿病KKAy小鼠有治疗作用。
Objective: To study the therapeutical effects of CW7213 on spontaneous non-insulin dependent diabetes(NIDD) KK Ay mice.Methods: The fasting blood glucose concentrations were tested at 0,1,2 and 3 h and 1,2,3 and 4 wk after the first doses of CW7213(20,60,120 μg.kg-1,4 weeks,qd).On the third week,intragastric loading glucose 2.5 g.kg-1 was given 15 min after s.c.CW7213,then the blood glucose concentrations were tested at 0.5,1,2 and 3 h.And the parameters including the fast blood serum insulin(INS),triglyceride(TG) and total cholesterol(TC) were tested at the end of the experiment.Results: The fasting blood glucose concentrations were reduced significantly at different times after administrating CW7213(P0.01,P0.05 vs control) and after loading glucose,the blood glucose concentrations were also reduced significantly.CW7213 could also remarkably increase the serum INS concentration and effectively reduce the serum TG concentration at the doses of 60 and 120 μg.kg-1.Conclusion: The results suggested that CW7213 could have therapeutical effects on KK Ay mice.
