摘要
目的观察NADPH氧化酶4(NOX4)在高肺血流肺动脉高压大鼠肺组织中表达的变化,探讨NOX4在高肺血流性肺动脉高压发病机制中的意义。方法 40只雄性SD大鼠随机分为对照组、分流4周组、分流8周组、分流11周组,通过腹主动脉-下腔静脉分流法建立高肺血流肺动脉高压大鼠模型,检测模型大鼠平均肺动脉压(mPAP)、右室肥厚指数(RVI),RT-PCR检测大鼠肺组织中NOX4 mRNA的表达情况。结果随着分流时间的延长,各分流组大鼠逐步出现了右室肥厚、平均肺动脉压升高等肺高压的表现,肺组织中NOX4 mRNA表达亦逐步增强。结论高肺血流肺动脉高压大鼠肺组织中存在着NOX4的显著表达,NOX4参与了高肺血流大鼠肺动脉高压的形成。
Objective To observe the expression of NOX4 in rats with pulmonary hypertension caused by high pulmonary blood flow and explore the implication of NOX4 in pathogenesis of left to right shunt pulmonary hypertension. Methods Forty male SD rats used were randomly divided into four groups in the experiment:control group,4 weeks group,8 weeks group and 11 weeks group. The model was established by abdominal aorta-inferior vena eava shunt. The mPAP, RVI of rats were measured and the expression of NOX4mRNA in rats were tested by RT-PCR. Results With the increasing of shunt time, right ventricular hypertrophy and elevation of mPAP in shunt groups gradually appeared. Meanwhile, the expression of NOX4mRNA was elevated gradually in rats. Conclusion The expression of NOX4 is enhanced obviously in lung tissue of rats with pulmonary hypertension caused by high pulmonary blood flow. NOX4 involves in formation of pulmonary hypertension in model rats.
