合成致死作用在抗肿瘤药物筛选中的研究进展

Advances in the study on synthetic lethal interactions in the screening of anticancer drugs

摘要 总结国外合成致死作用在抗肿瘤药物筛选中的最新研究进展。采用PubMed数据库检索系统,以"合成致死作用、抗肿瘤、药物筛选"等为关键词,检索1990年至今合成致死作用在抗肿瘤药物发现中的作用的文献。合成致死作用研究主要集中在PARP抑制剂和BRCA1/2缺失、MSH和DNA聚合酶、VHL基因、PTEN基因缺失、RAS和MYC基因的失调等。合成致死作用在抗肿瘤药物筛选中具有广泛应用,有望作为新一代抗肿瘤药物的筛选方法。 The aim of this review is to summarize the research advances in the role of synthetic lethal interactions in anticancer drug screening.Based on the key words such as synthetic lethal interactions,anti-cancer and drug screening,we searched the publications related to the discovery of anticancer drugs based on the synthetic lethal interactions for 20 years(Jan 1,1990-May 3,2011) from PubMed.The previous studies indicated that synthetic lethal interactions in the discovery of anticancer drugs have mainly been focused on PARP inhibitors and BRCA1/2 deficiency,MSH and DNA polymerases,VHL,the loss of PTEN,dysregulation of RAS and MYC.Synthetic lethal screening of anticancer drugs is widely applied,and it is a promising method for screening new generations of anticancer drugs.
