太湖水体中藻蓝蛋白提取的影响因素分析 被引量:1

摘要 以室内培养的铜绿微囊藻样品为参照,基于反复冻融法,探讨了滤膜类型、磷酸盐缓冲液加入体积、冻融次数对太湖水体中藻蓝蛋白提取的影响。结果表明:1)GF/C玻璃纤维滤膜(孔径1.2μm)和CN-CA混合纤维素酯微孔滤膜(孔径0.45μm)提取的叶绿素a结果差异不显著(p分别为0.95和0.97),说明二者的生物截留量基本一致;2)在生物截留量相当的情况下,GF/C滤膜和CN-CA滤膜提取的藻蓝蛋白结果差异显著(p均为0.04),GF/C滤膜的提取效率高于CN-CA滤膜;3)加入的缓冲液体积和藻蓝蛋白质量浓度呈良好的负线性关系(r均大于0.85),受藻体包裹效应的影响,太湖水体对比组中的相关系数略低于人工培养的铜绿微囊藻;4)太湖水样和铜绿微囊藻在冻融次数上存在差异,太湖水样冻融9~11次的提取效果较佳,室内培养的铜绿微囊藻冻融次数以5~7次为宜。 The present paper intends to introduce our experimental analysis results of the influential factors of the phycocyanin extraction from the water samples in Taihu Lake,Jiangsu.For our research purpose,we have collected samples from Meiliang Bay of the lake,in August 20,2011,and accomplished our analysis through the contrast experiment by taking Microcystic aeruginosa indoor-cultured as reference and based on the freezing-thawing cycle.The influential factors we refer to include filter type,phosphate buffer solution(PBS) added concentration,and the number of freezing and thawing cycle.The results of our analysis indicate that:(1) Using the chlorophyll A concentration as an evaluation index,we have made a comparison between the retentate capacity of the filtration aperture of GF/C glass fiber filters(GF/C) and CN-CA micropore membrane filters(CNCA) so as to improve our analysis of the difference in the phycocyanin extraction from the lake water samples.The results of variation analysis prove that the difference between GF/C and CN-CA in chlorophyll A extraction of the water samples from the Lake and the algal samples cultured indoor proves insignificant(p=0.95 and 0.97,respectively).Actually,the filtration aperture of the GF/C and CNCA has had minus influence on the retentate capacity.(2) However,for the same creature retentate capacity,the difference between GF/C and CN-CA in extracted water samples from the Lake and algal samples turns to be significant(p=0.04).The extraction efficiency of phycocyanin in GF/C turns to be higher than that in CN-CA,and therefore GF/C should be recommended to extracted from the lake as water samples.(3) There were significant linear negative correlation between the PBS addition concentration and phycocyanin concentration in the water samples and the algal samples,with the correlation coefficient of each group being over 0.85.Affected by the algae package(when the algal gathered together),the correlation coefficient in water samples from Taihu Lake proves to be lower than that in Microcystic aeruginosa indoor-cultured.(4) The length of freezing and thawing cycle between the natural water samples and algal samples turns to be different.The available freezing circles tend to be 9 to 11 times for natural water samples,and 5 to 7 times for Microcystic aeruginosa.
