Effects of Regulatory Dendritic Cells on the Airway Inflammation and Expression of IL-12 and IgE in Mice with Asthma

摘要 目的用肺脏基质细胞培养上清液与成熟树突状细胞(mDC)共培养得到的调节性树突状细胞(rDC)回输小鼠,观察其对哮喘小鼠气道炎症的抑制作用及对白介素12(IL-12)、IgE表达的影响。探讨rDC诱导免疫耐受的可能机制,为临床防治哮喘提供新的思路。方法制备C57BL/6小鼠rDC。将40只C57BL/6小鼠分4组,每组10只。分别为正常组、卵清蛋白(OVA)组、mDC组、rDC组。第21天处理全部小鼠,观察各组小鼠肺组织病理学改变,血清及肺泡灌洗液中IL-12及血清IgE的表达。结果 rDC回输后可减轻肺部炎症表现,4组小鼠血清中IL-12、IgE比较,差异均有统计学意义(P<0.01)。其中rDC组血清中IL-12的水平高于OVA组,IgE低于OVA组,差异均有统计学意义(P<0.01)。结论回输rDC可在一定程度上缓解哮喘小鼠气道的过敏性炎症反应,与上调Th1型细胞因子IL-12、下调IgE的表达有关。 Objective To investigate the effects of regulatory dendritic cells co - cultured by supernatant of pulmonary stromal cells and mature dendritic cells (mDC) on the airway inflammation and interleukin (IL) -12 and IgE in murine model of asthma and to elucidate the possible mechanism of regulatory dendritic ceils (rDC) in the induction of immune tolerance in or- der to provide new thoughts for the clinical prevention and treatment of asthma. Methods Regulatory dendritic cells of C57BL/6 mice were prepared. 40 C57BL/6 mice were divided into four groups (normal group, OVA group, mDC group and rDC group) with each group 10 mice. On day 21, all mice were disposed. Pathological changes in pulmonary tissues of each group and expres- sion of IL - 12 and IgE in serum and in bronchoalveolar lavage fluid were observed. Results The reinfusion of rDC could alleviate lung inflammation. The differences of serum IL - 12 and IgE were statistically significant among the four groups ( P 〈0. O1 ). Ser- um IL - 12 level of rDC group was significantly higher than that of the OVA group, while IgE level was significantly lower than that of the OVA group (P 〈 0.01 ) . Conclusion Reinfusion of regulatory dendritic cells can to some extent alleviate airway in- flammation in mice models with asthma and this may relate to the up - regulation of Thl type cytokine interleukin (IL) - 12 and down - regulation of IgE.
