摘要
目的从兔骨骼肌中提取肌球蛋白轻链,为下一步研究肌肉收缩原理提供可磷酸化的底物。方法用离心、透析、柱层析、SDS聚丙烯酰胺凝胶电泳(SDS-PAGE)和电泳转移等方法纯化兔骨骼肌肌球蛋白轻链。结果提取纯化得到较纯的肌球蛋白,进一步得到轻链混合液和各种轻链纯品,经紫外吸收检测及SDS0PAG证实其分子量分别为25000(MLC1)、19000(MLC2)和16500(MLC3),均与文献报道基本一致。结论研究所用的方法可行,所得到的肌球蛋白及其轻链样品较纯,可用于下一步实验。
Objetive TO obtain myosin light chains (MLC) from rabbit skeletal muscle in order to providephosphorylatable substrate for muscle contraction. Mithnds The centrifugation, dialsis, ion exchange chro-matOgraphy, SDSPAGE and electrophoresis transferring were used to obtain MLC. Results Pure myosins andall kinds of light chains were obtained and their behaviors are similar with those references reported. Theirmolecular weight are 25000(MLC), 19000(MLC2) and 16500(MLC3) respectively. Conclusion The methodsare practicable and the purified MLC can be used fOr further study.
