摘要
目的:研究COPD大鼠骨骼中磷酸化NF-κB通路蛋白的表达情况及意义。方法:采用气管内滴入脂多糖+反复熏香烟法复制COPD大鼠动物模型。Western blot方法测定大鼠骨骼肌内磷酸化NF-κB蛋白的表达,ELISA法测定大鼠血清及膈肌中IL-1β的表达。结果:Western blot结果显示,COPD大鼠骨骼肌中磷酸化NF-κB蛋白的表达均较对照组明显升高(P<0.01);ELISA结果显示,IL-1β在血清及骨骼肌中的表达均升高(P<0.01);NF-κB与IL-1β水平呈显著正相关(r=0.55,P<0.01)。结论:COPD大鼠骨骼肌中NF-κB表达上调,IL-1β可激活骨骼肌中NF-κB通路。
Objective: To study the role of NF-κB pathway in the skeletal muscle of COPD rats. Methods: The COPD rat model was established by instillation of LPS and exposure to cigarette smoke for 28 days. The protein levels of NF- K B in the skeletal muscle were measured by Western blot. The concentration of IL-1 β in serum and diaphragm were measured by ELISA. Results: Western blot resuit showed that, in the skeletal muscle, compared with the normal control group, the protein expression of NF-K B increased significantly in the model group(P〈0.01). Compared with the normal control group, in the serum and the diaphragm, the IL-1βwere significantly higher than those of the control group(P〈0.01). NF- κB was significantly positively correlated with the IL-1β (r=0.55, P〈0.01). Conclusion: The data shows that the NF-K B pathway is up-regulated in the skeletal muscle of COPD rats, and IL-1β may activate NF- κB pathway in skeletal muscle.
