摘要
目的 :建立测定血浆中异烟肼浓度的反相高效液相色谱法。方法 :采用C18BDS柱 ,流动相为 0 .0 2mol·L-1磷酸二氢钾∶乙腈 (6 9∶31)。 2 0 0 μl血浆用 2 0 %三氯醋酸沉淀蛋白后 ,上清液加桂皮醛衍生化后直接进样。 结果 :本方法在 0 .1~ 13.88μg·ml-1范围内线性良好。日内和日间误差均小于 10 % ,方法回收率 >95 %。结论 :此方法具有操作简便、灵敏度较高、无干扰等特点 。
OBJECTIVE:An RP HPLC method has been developed to determine the concentration of isonizid in plasma.METHODS:The C 18 column was used.The mobile phase was 0.02 mol·L -1 KH 2PO 4∶acetonitrile(69∶31).The plasma sample was injected directly for determination after being deproteinized with 20% trichloroacetic acid and reacted with cinnamaldehyde.RESULTS:This method showed good Linear relationship from 0.1 ~ 13.88 μg·ml -1 .The intraday precision and interday precision were less than 10%.The recovery of the method was more than 95%.CONCLUSIONS:This method is simple,sensitive and specific.This method has been used in pharmacokinetics and bioavaiability study of compound rifampicin tablets.
